Research Interests

The Gebregiworgis (Gebre) lab is interested in understanding mechanistic details of deregulated cell signaling and metabolism. Our first area of research interest focuses on the molecular mechanisms of small GTPase protein functions. GTPase signaling is altered in multiple cancers and developmental diseases. Many small GTPases, such as RAS, are prime targets for cancer drug discovery. As a result, biochemical and structural understanding of small GTPase regulation and interaction with effectors and regulators is paramount. In this avenue, our lab is interested in exploring the biochemical, structural, and cellular impact of posttranslational modifications and mutations of other small GTPases. We employ various structural, biochemical, biophysical, and molecular tools for our investigation.

Our second area of research is developing and applying NMR metabolomics-based methods to investigate disease-associated metabolic alterations. Metabolomics is a systemic and comprehensive analysis of metabolites in biological specimens. Metabolites are ultimately products of gene expression; thus, metabolite profiling provides phenotypic data that are the downstream output of genetics, transcriptomics, and proteomics. As a result, altered metabolic profiles provide proximate information about the phenotype under investigation. In this regard, we are interested in studying the impact of metabolite crosstalk between compartments (e.g., cancer and stromal cells; microbiome flora and human cells) and employing metabolomics to promote precision medicine.
